Effects of Neutrophil Elastase

چکیده
A bstract. The effects of neutrophil elastase on endothelial prostacyclin (PGI2) production, nucleotide release, and responsiveness to vasoactive agents were compared with the effects of cathepsin G (the other major neutral protease of neutrophils), pancreatic elastase, trypsin, chymotrypsin, and thrombin. PGI2 production by pig aortic endothelial cells cultured on microcarrier beads and perfused in columns was stimulated in a dose-dependent manner by trypsin, chymotrypsin, and cathepsin G (1-100 ,ug/ml for 3 min). Thrombin, while active at low concentrations (0.1-10 National Institutes of Health U/ml), induced smaller responses. Neutrophil and pancreatic elastase had little or no effect on PG12 production. Dose-dependent, selective release ofadenine nucleotides was induced by neutrophil elastase (3-30 .g/ml). The other proteases were much less active; for example, trypsin (100 tg/ml) induced a response only -5% as great as did 30 ,ug/ml neutrophil elastase. After exposure to 30 ug/ml neutrophil elastase, cells did not exhibit the characteristic burst of PG12 production in response to extracellular ATP; responsiveness gradually returned after 40-120 min. This effect was not seen with the other proteases. Elastase partly inhibited re-

In Vitro Inhibition of Neutrophil Elastase Activity by Inhaled Anti-Pseudomonas Antibiotics Used in Cystic Fibrosis Patients
BACKGROUND Inhaled antibiotics are commonly used in the treatment of cystic fibrosis lung disease. A previous study suggested neutrophil elastase activation by colistin in vitro. Here, we investigated direct effects of the commonly used antibiotics colistin and tobramycin on neutrophil elastase activity. METHODS Neutrophil elastase was measured spectrophotometrically. The antibiotics colistin...
متن کاملEffect of nebulised recombinant DNase on neutrophil elastase load in cystic fibrosis.
BACKGROUND DNA released by degenerating inflammatory neutrophils contributes to mucous plugging of airways in patients with cystic fibrosis. Neutrophil elastase, a major effector of tissue destruction in the lungs of patients with cystic fibrosis, is a highly cationic molecule which is bound and inhibited by negatively charged polyanions such as mucin and DNA in purulent sputum. Thus, the solub...
متن کاملSivelestat reduces inflammatory mediators and preserves neutrophil deformability during simulated extracorporeal circulation.
BACKGROUND Neutrophil is a major focus in efforts to ameliorate the systemic inflammatory response associated with cardiopulmonary bypass. Neutrophil elastase is a powerful proteolytic enzyme, and plays a pivotal role in the development of the inflammatory response. This study assesses the inhibitory effects of sivelestat, a highly specific neutrophil elastase inhibitor, on elastase levels, cyt...
متن کاملNeutrophil Elastase Cleaves C3bi on Opsonized Pseudomonas
Neutrophil elastase has been implicated as a factor that impairs local host defenses in chronic Pseudomonas aeruginosa (Pa) lung infection in cystic fibrosis (CF). We recently showed that this enzyme cleaves the C3b receptor, CR1, from neutrophils (PMN) in the lungs of infected CF patients. The C3bi receptor on these cells, CR3, is resistant to elastase. We now show that purified neutrophil ela...
متن کاملRelative α1-anti-trypsin deficiency in systemic sclerosis
OBJECTIVE Neutrophil elastase is secreted by neutrophils during activation and circulates in the plasma where it can play a role in inflammation and fibrosis. This study examines the role of neutrophil elastase in SSc, a systemic CTD that is typified by vascular dysfunction, tissue fibrosis and inflammation. METHODS Serum neutrophil elastase and α₁-anti-trypsin concentrations were assessed in...
